No. What you have in the defined View is what will display on the Layout.

<T>I have a guideline layer which I would like to keep hidden but seems to appear on the final layout even though the original has that particular layer hidden.

Is there a way to hide or show layers in layout without effecting the original? and vice versa

Sujey</T>

Two layer combinations, one with guidelines on for working, one with guidelines off which is assigned to the view that is placed on the layout.
