Hi there,

I would like to know how to subtract the internal fill from the external fill please, so you're just left with the blue area as a fill.

Select your fill, select the fill tool and draw inside your big one. And it will substract a hole. 

And to substract an existing fill/ shape, you can use the magic wand (default shortcut: spacebar) to select it.

I suggest you turn on Tool Tips and have a quick look at the different pet pallette options you have for the different tools as you come across them. With all of the basic questions, you will probably save a bit of time over waiting for a reply each time.
